Athletes will run past some of the city's main attractions, crossing the Clyde four times before finishing in Glasgow Green.
Commonwealth Games bosses, with Scottish marathon star Derek Hawkins, unveiled the route under the McLellan Arch in the Merchant City park on Friday.
The two-lap 26.2 mile route passes through Merchant City, George Square, Buchanan Street and the Clyde Arc, as well as Bellahouston Park and Pollok Park.
Fans will be able to line the streets for free to cheer on their running heroes.
The marathon medal ceremony will be held in Hampden Stadium following the event on Sunday, July 27.
David Grevemberg, Glasgow 2014 chief executive, said: "The Glasgow 2014 Marathon is a wonderful showcase event for the Games and one that everyone can come and enjoy.
"The route the athletes will take really shows Glasgow at its best and will bring the spectacle of live competitive sport right into the heart of the city."
