Shettleston MSP Frank McAveety is a keen Celtic fan while Calton councillor George Redmond is a huge Rangers supporter.
Both are running in the third East End 5K, which is being held this Sunday.
They have agreed to swap football tops for the run - but only if they get donations which will go to a local charity.
Mr Redmond said: "This is my worst nightmare - I cannot contemplate putting on a Celtic jersey but will do so if we can raise money for a local volunteer group.
"It is important we recognise the work going on in the local area because we are honoured to have such a fantastic network of voluntary organisations in the East End.
"Anything we can do to support them, we will, even if it means we have to do something as extreme as this."
Last year Mr Redmond shocked Mr McAveety when he said they would donate £1 to charity for everyone who beat them in the 5K.
The councillor accused his MSP chum of being "as tight as two coats of paint" but between them they donated £500 to a local group.
This year the competition between the friends is as fierce as ever, with Mr Redmond convinced he will cross the line first.
The race starts at Tollcross Park at 10am.
