The Atlantic Quay project will see two office buildings and a six-storey, 150-bedroom hotel built on the site between James Watt Street and York Street.
A public square will also be created along with shops and leisure facilities and developers hope to include a health club and conference centre.
The plan, lodged with Glasgow City Council by the Capella Group, is a key part of the regeneration of the former Broomielaw area -now known as the Inter-national Financial Services District.
Dubbed "Wall Street on the Clyde" it has attracted 15,000 jobs and major financial institutions like Barclays, Morgan Stanley and Direct Line since it was launched in 2001.
The chief executive of the Glasgow-based Capella Group said he wanted to create a "heart" for the district, formerly made up of warehouses and built around Broomielaw Quay, the traditional departure point for Clyde and Irish steamers.
The company is retaining the A-Listed warehouse facade on James Watt Street, built in the mid-19th century.
In 1968 the street was the scene of a major tragedy when 22 people died in a fire in an upholstery factory.
Chief executive Jim Fitzsimons said: "At a time when the economic climate is so challenging we see this project as a major vote of confidence in the Broomie-law area and a timely boost for Glasgow's commercial property sector.
"We believe the city is far better placed than most other UK cities to quickly come out of the current downturn, and believe that there will be significant take up of office space this year."

"The IFSD initiative has been tremendously successful in attracting thousands of jobs and many millions of pounds of investment to Glasgow and the addition of high quality office space at Atlantic Square will further enhance its profile."
The two office buildings will total 227,000 sq ft and there will be a covered courtyard, which developers see as a key feature.
Glasgow City Council has already granted outline planning permission for a number of elements of the project and Capella Group is now seeking approval for a revised scheme.
